RESUMO

Abstract The objective of this work was to perform the phytochemical characterization, to determine total phenols, antioxidant (AAO%) and antimicrobial potential of the ethanolic extracts of carambola. The phytochemical study was carried out through a qualitative analysis of the chemical constituents and quantitative determination of the phenol content By the Folin-Ciocalteu test. Qualitative and quantitative antioxidant tests were performed using the DPPH method (2,2 diphenyl-1-picryl-hydrazila) and iron reduction (FRAP). The minimum inhibitory concentration (MIC) was determined by microdilution in 96-well plates. The presence of pyrogallic tannins, steroids and saponins has been identified. The highest total phenol content, quantified in the samples, was found in the stem bark (0.0866 mgEAG/g) and in the fruit (0.0734 mgEAG/g). In the antioxidant evaluation, the extracts of the green fruit bagasse (AAO% 71.9%,) and stem bark at 50 μg/mL (AAO% 94%) with CE50 23.7 μg/mL. Leaf extracts, stem bark, ripe fruit bagasse and green fruit bagasse presented MICs of 100 μg/mL against multiresistant pathogenic bacteria and fungi.

RESUMO

Genotypes x environment (G x E) interaction consists of different behavior of genotypes cultivated in different environments. This interaction occurs due to the performance variation of each genotype in different environments. To reduce the effect of the interaction in soybean crops, some studies have been reported in the literature to study their adaptability and stability. However, these studies are still scarce in Minas Gerais State. Thus, the aim of this study was to verify the adaptability and stability of soybean cultivars and identify the cultivars that contribute least to the G x E interaction in Minas Gerais. Six soybean cultivars were evaluated in 9 different environments. The plots were composed of 4 rows of 5 m with a spacing of 0.5 m between rows, and only the two central rows were harvested. The inoculation with Bradyrhizobium japonicum was performed through sowing furrow. The fertilization followed the recommendations of the Soil Fertility Commission of Minas Gerais. Grain yield was evaluated in kg/ha after conversion to 13% moisture. After individual analysis, the joint analysis was performed by grouping the phenotypic means by the Scott and Knott (1974) test. Wricke's ecovalence methodologies and the Annicchiarico confidence index were applied for the adaptability and stability analysis. The interaction was decomposed into a simple and a complex part. The cultivars BRSMG 820RR and BRSMG 760SRR have wide adaptability and stability. The first one presents a better index of confidence and a small contribution to the interaction.

RESUMO

This study aimed at verifying the adaptability and stability of soybean cultivars, considering the grain yield and quality of seeds, adopting univariate and multivariate approaches. The experiments were conducted in two crops, three environments, in 2013/2014 and 2014/2015 crop seasons, in the county of Inconfidentes, Lavras, and Patos de Minas, in the Minas Gerais State, Brazil. We evaluated 17 commercial soybean cultivars. For adaptability and stability evaluations, the Graphic and GGE biplot methods were employed. Previously, a selection index was estimated based on the sum of the standardized variables (Z index). The data relative to grain yield, mass of one thousand grain, uniformity test (sieve retention), and germination test were standardized (Zij) per cultivar. With the sum of Zij, we obtained the selection index for the four traits evaluated together. In the Graphic method evaluation, cultivars NA 7200 RR and CD 2737 RR presented the highest values for selection index Z. By the GGE biplot method, we verified that cultivar NA 7200 RR presented greater stability in both univariate evaluations, for grain yield, and for selection index Z.

RESUMO

The aim of this study was to verify the adaptability and stability of soybean cultivars with regards to yield and oil content. Data of soybean yield and oil content were used from experiments set up in six environments in the 2011/12 and 2012/13 crop seasons in the municipalities of Patos de Minas, Uberaba, Lavras, and São Gotardo, Minas Gerais, Brazil, testing 36 commercial soybean cultivars of both conventional and transgenic varieties. The Wricke method and GGE biplot analysis were used to evaluate adaptability and stability of these cultivars. Large variations were observed in grain yield in relation to the different environments studied, showing that these materials are adaptable. The cultivars exhibited significant differences in oil content. The cultivars BRSGO204 (Goiânia) and BRSMG (Garantia) exhibited the greatest average grain yield in the different environments studied, and the cultivar BRSMG 760 SRR had the greatest oil content among the cultivars evaluated. Ecovalence was adopted to identify the most stable cultivars, and the estimates were nearly uniform both for grain yield and oil content, showing a variation of 0.07 and 0.01%, respectively. The GGE biplot was efficient at identifying cultivars with high adaptability and phenotype stability.

RESUMO

Bothrops insularis is an endemic and critically endangered snake with an estimated population of 2000 individuals restricted to Queimada Grande Island, in southeastern Brazil. Brazilian researchers established a captive breeding program for the species that includes the application of assisted reproductive technologies. The present study, therefore, aimed to evaluate semen samples from captive B. insularis throughout the year to ascertain seasonal differences in semen traits as well as correlations with body size and weight. Eighteen males with snout-vent length (SVL) ranging from 43.5 to 73.7 cm were collected at quarterly basis between August 2012 and May 2013. Macroscopic analysis revealed semen volumes ranging from 0.5 to 6.0 µL with samples featuring whitish to yellowish color and creamy and thick consistency. Viable sperm was obtained from all males indicating that individuals with SVL equal to or greater than 43.5 cm are sexually developed. However, adult and immature males (estimated by SVL) exhibited different seasonal profiles for motility and progressive motility. Adult males had a decrease in sperm motility and progressive motility during summer and spring, respectively, whereas the same variables did not vary throughout the year in immature snakes. Sperm concentration in all individuals was less (0.5 × 10(9) µL) during the winter, but no seasonal fluctuations were detected in semen volume. These findings are of particular importance to the development of reproductive tools such as male selection, artificial insemination and sperm freezing for the genetic management of this critically endangered snake.

RESUMO

Soybean is one of the main sources of foreign exchange credits for Brazil in the agricultural sector. There is increasing interest in growing this leguminous crop, especially in the southern region of Minas Gerais, due to its importance as an alternative for crop rotation with maize. In this respect, the study of the adaptability of new cultivars to the region is indispensable so as to obtain high yields. Thus, the aim of this study was to evaluate the performance of 38 soybean cultivars for growing in the summer season in the municipality of Lavras, MG, Brazil, in the 2010/2011 and 2011/2012 crop years. The experiments were conducted in a randomized block design with 3 replications and the treatments consisted of 38 cultivars. At the time of harvest, the following assessments were made: grain yield (kg/ha), height of the lowest pod (cm), plant height (cm), and lodging. The data were subjected to individual and combined analysis of variance. The phenotypic mean values were clustered, adopting the Scott and Knott test. For simultaneous selection of multiple traits, the sum of rank index of Mulamba and Mock was adopted. The cultivar TMG 801 RR had the best yield performance; the cultivars Monsoy 8001, MGBR-46 (Conquista), and BRSMG 68 (Vencedora) also stood out. Considering simultaneous selection for grain yield, plant height, height of the lowest pod, and lodging, the cultivar TMG 801 RR is recommended for growing in the summer season in the southern region of Minas Gerais.

RESUMO

BACKGROUND: Medical equipment coming into contact with non-intact skin or mucous membranes is classified as semi-critical material. This equipment requires at least high-level disinfection, as the major risk in all invasive procedures is the introduction of pathogenic microbes causing hospital-associated infections. AIM: To evaluate the capacity of ozone gas and ultrasound to disinfect semi-critical, thermally sensitive material. METHODS: Used corrugated tubing from mechanically ventilated tracheostomized patients in the intensive care unit was obtained. Enzymatic detergent was applied for 15min before different disinfection techniques were evaluated as follows: Group A (0.2% peracetic acid); Group B (ultrasound for 60min); Group C (application of ozone gas at a concentration of 33mg/L for 15min); Group D (ultrasound for 30min and ozone for 15min); Group E (ultrasound for 60min and ozone for 15min). FINDINGS: Application of ultrasound for 60min reduced the level of microbial contamination by 4 log10, whereas ozone alone and the other two combined techniques (ultrasound and ozone) and the peracetic acid reduced the level of microbial contamination by 5 log10. CONCLUSION: Ozone was the most advantageous technique taking into consideration processing time, ease of use, effectiveness, and cost. The use of ozone gas to disinfect semi-critical material proved to be technically feasible and extremely promising.

This work aimed to evaluate the methodology of tetrazolium test in Amburana cearensis (Allemão) A.C. Smith seeds, establishing the best pre-moistening conditions, as well as temperature, coloration period and tetrazolium solution concentration. The coloration of intact and scarified (sanded) seeds, both with tegument, was directly evaluated in tetrazolium solution at the concentrations of 0.025; 0.050; 0.075; and 1 percent, at 35 and 40ºC, for 12 and 24 h in the dark. Pre-moistening to remove the tegument was evaluated by using intact and scarified seeds, which were directly immersed in water and subjected to imbibition in paper towel for 12, 24, 36, and 48 h at 35 and 40ºC. After establishing the method to remove the tegument, embryos were subjected to coloration in tetrazolium solution at the concentrations of 0.025; 0.050; 0.075; and 1 percent. Based on the obtained results, the complete tegument removal is needed for coloration; besides, the temperature of 40ºC is harmful for imbibition during pre-moistening. The tetrazolium test in A. cearensis seeds should be performed including pre-moistening through direct immersion of scarified (sanded) seeds in water for 24h at 35ºC for later complete tegument removal. To achieve the ideal coloration, embryos must be immersed in tetrazolium solution at 0.05 percent for 3h at 40ºC.

RESUMO

The treatment of neurological disorders and neurodegenerative diseases is related to the levels of acetylcholine (ACh) through the inhibition of acetylcholinesterase (AChE). Galanthamine, an important alkaloid isolated from the Amaryllidaceae family, is approved for the pharmacological treatment of Alzheimer's disease (AD) and acts by inhibiting the acetylcholinesterase (AChE) activity. In the present study, Ellman's method was used to verify the inhibition of AChE activity of some isoquinolines alkaloids such as galanthamine, montanine, hippeastrine and pretazettine. At the concentrations 1mM, 500 microm and 100 microm, galanthamine presented an AChE inhibition higher than 90%. Montanine inhibited, in a dose-dependent manner, more than 50% of the enzyme at 1mM concentration. With the concentrations 500 microm and 100 microm, 30-45% of AChE activity inhibition was detected. The alkaloids hippeastrine and pretazettine presented no significant inhibition of the AChE activity. The results demonstrate that montanine significantly inhibits AChE activity at the tested concentrations, suggesting the necessity of further investigations on this alkaloid use in treating neurological disorders.

RESUMO

Ascidians are urochordates, marine invertebrates with non-feeding motile chordate tadpole larvae, except in the family Molgulidae. Urodele, or tailed, Molgulids have typical ascidian chordate tadpole larvae possessing tails with muscle cells, a notochord, and a dorsal hollow nerve cord. In contrast, anural (or tail-less) Molgulids lack a tail and defining chordate features. Molecular phylogenies generated with 18S and 28S ribosomal sequences indicate that Molgulid species fall into at least four distinct clades, three of which have multiple anural members. This refined and expanded phylogeny allows careful examination of the factors that may have influenced the evolution of tail-less ascidians.
